      为了探讨南瓜色素的制备工艺,并研究其清除自由基的能力与在线虫体内的抗氧化作 用。采用超声波辅助有机溶剂工艺提取南瓜色素,并通过Box-Behnken Design(BBD)的试验设计 优化提取条件;大孔吸附树脂纯化后,研究南瓜色素体外自由基的清除能力与线虫体内的抗氧化 效果。结果显示,超声提取参数为超声时间42 min、浸提时间80 min、料液比1∶66时,南瓜色素得 率为6.760 ± 0.005 mg/g。经大孔树脂对南瓜色素静态和动态吸附,AB-8为最佳型号,静态吸附和 解吸时间分别是150 min和50 min,树脂与溶剂最佳比例为1∶8(g/mL),静态吸附pH值为6,动态吸 附洗脱剂的最佳用量为 18 倍柱体积。南瓜色素可显著提高线虫体内的抗氧化物酶(SOD)活性 (P < 0.05),降低MDA水平(P < 0.05)。

      In order to study the preparing conditions of pumpkin pigment, the ability of scavenging free radicals and its an⁃ tioxidative effect in caenorhabditis elegans, the pure pumpkin pigment was prepared in this experiment. The pumpkin pig⁃ ment was prepared by ultrasonic assisted organic solvent extract and box Behnken design (BBD). After purification with macroporous resin, the scavenging ability of free radicals in vitro of pumpkin pigment and the antioxidant effect in vivo of C. elegans were studied.The extraction yield of pumpkin pigment was 6.760 ± 0.005 mg/g under the optimized conditions: ul⁃ trasound time of 42 min, extraction time of 80 min and solid-liquid ratio of 1:66. According to static and dynamic adsorp⁃ tion of pumpkin carotenes with macroporous resin, AB-8 was the best type. The static adsorption time were 150 min and the resolution time is 50 min. The optimum ratio of resin to solvent was 1:8 (g/mL), the static adsorption pH was 6, and the optimum dosage of dynamic adsorption eluent was 18 column volume. Besides, pumpkin pigment could significantly in⁃ crease SOD activity (P < 0.05) and decreased MDA level (P < 0.05) in C. elegans.
